现在准备开始办perm，遇到一个问题: 我是去年7月开始工作的，8月才拿到正式的学位证，但是6月就已经答完辨。现在写job description的时候能够写要求master吗？（我当时申请EB1用的是本科学位）

IF you already applied EB1, why PERM now?

According to my understanding, the position for PERM can be a new position different from your current one and therefore you should be able to list Master degree as a requirement for this PERM position. And you will get a "Promotion" for this PERM position. However, I would highly recommend you to consult your lawyer.
