What is the group factor, treatment or block? What is the subgroup factor? Is subgroup factor independent of group factor or nested within it? If it is the former, then it seems you have a 2x2 factorial design. If it is the latter, then it is a block design. For either case, you should use ANOVA to evaluate the differences between groups or subgroups. However, ANOVA is just a general term, there are different methodologies for different designs. Personally, I don't think two-sample t test or Wilcoxon rank sum test will do the work in this case. Just my two cents.
